They arrived in the southern colonies and settled along the Elizabeth, Potomac, and James Rivers. By the 1690s they had organized a church in Norfolk. They also settled in Maryland and Delaware around the Chesapeake Bay. A considerable number of Presbyterians settled in New England between the 1625s and 1640s, although they were sometimes congregationalized in those early years. Those who settled in Connecticut maintained their Presbyterian sympathies. Some Presbyterians immigrated to the Carolinas and established congregations in Charleston as early as the seventeenth century.

It was formed to meet the needs of those people in the "desolate places" of the American colonies. Moreover, presbytery members represented a multicultural mix from the very beginning, albeit all from the British Isles: John Andrews was from England; John Wilson and A. N. Taylor were from New England; George McNish came from Scotland; and John Hampton along with Samuel Davis represented the Scotch-Irish. Francis Makemie also was from Ireland, and Welsh pastors Howell Powell and Malachi Jones soon associated with the organization.
